Polypeptide Multilayer Self-Assembly Studied by Ellipsometry
نویسندگان
چکیده
A polypeptide nanofilm made by layer-by-layer (LbL) self-assembly was built on a surface that mimics nonwoven, a material commonly used in wound dressings. Poly-L-lysine (PLL) and poly-L-glutamic acid (PLGA) are the building blocks of the nanofilm, which is intended as an enzymatically degradable lid for release of bactericides to chronic wounds. Chronic wounds often carry infection originating from bacteria such as Staphylococcus aureus and a release system triggered by the degree of infection is of interest. The dry nanofilm was studied with ellipsometry. The thickness of the nanofilm was 60% less in its dry state than in its wet state. The measurements showed that a primer was not necessary to build a stable nanofilm, which is practically important in our case because a nondegradable primer is highly unwanted in a wound care dressing. Added V8 (glutamyl endopeptidase) enzymes only showed adsorption on the nanofilm at room temperature, indicating that the PLL/PLGA "lid" may remain intact until the dressing has been filled with wound exudate at the elevated temperature typical of that of the wound.

Control of stability of polypeptide multilayer nanofilms by quantitative control of disulfide bond formation
The crosslinking of polymers in a polymeric material will alter the mechanical properties of the material. Control over the mechanical properties of polyelectrolyte multilayer films (PEMs) could be useful for applications of the technology in medicine and other areas. Disulfide bonds are ‘natural’ polypeptide crosslinks found widely in wild-type proteins. Here, we have designed and synthesized ...
متن کاملTunable drug loading and release from polypeptide multilayer nanofilms
Polypeptide multilayer nanofilms were prepared using electrostatic layer-by-layer self-assembly nanotechnology. Small charged drug molecules (eg, cefazolin, gentamicin, and methylene blue) were loaded in polypeptide multilayer nanofilms. Their loading and release were found to be pH-dependent and could also be controlled by changing the number of film layers and drug incubation time, and applyi...
متن کاملMass Transfer in Amperometric Biosensors Based on Nanocomposite Thin Films of Redox Polymers and Oxidoreductases
Mass transfer in nanocomposite hydrogel thin films consisting of alternating layers of an organometallic redox polymer (RP) and oxidoreductase enzymes was investigated. Multilayer nanostructures were fabricated on gold surfaces by the deposition of an anionic self-assembled monolayer of 11-mercaptoundecanoic acid, followed by the electrostatic binding of a cationic redox polymer, poly[vinylpyri...
متن کاملBiocompatible polypeptide microcapsules via templating mesoporous silica spheres.
We reported the stepwise formation of biocompatible poly(L-lysine)/poly(L-glutamic acid) (PLL/PGA) multilayer films on mesoporous silica (MS) spheres via layer-by-layer (LbL) self-assembly technique. In-situ QCM revealed the nonlinear (exponential) growth of PLL/PGA multilayer films at both pH 5.5 and pH 7.0 conditions. Xi-potential measurements of the multilayer coated particles indicated that...
متن کاملThe structure of self-assembled multilayers with polyoxometalate nanoclusters.
Using electrostatic layer-by-layer self-assembly (ELSA), the formation of multilayers with polyelectrolytes and nanoscopic polyoxometalate (POM) clusters of different sizes and charges is investigated. The multilayers are characterized by UV-vis absorption spectroscopy, optical ellipsometry, cyclic voltammetry, and atomic force microscopy. In all cases, it is possible to find experimental condi...
